    Downregulation of Brassica napus MYB69 (BnMYB69) increases biomass growth and disease susceptibility via remodeling phytohormone, chlorophyll, shikimate and lignin levels

    MYB transcription factors are major actors regulating plant development and adaptability. Brassica napus is a staple oil crop and is hampered by lodging and diseases. Here, four B. napus MYB69 (BnMYB69s) genes were cloned and functionally characterized. They were dominantly expressed in stems during lignification. BnMYB69 RNA interference (BnMYB69i) plants showed considerable changes in morphology, anatomy, metabolism and gene expression. Stem diameter, leaves, roots and total biomass were distinctly larger, but plant height was significantly reduced. Contents of lignin, cellulose and protopectin in stems were significantly reduced, accompanied with decrease in bending resistance and Sclerotinia sclerotiorum resistance. Anatomical detection observed perturbation in vascular and fiber differentiation in stems, but promotion in parenchyma growth, accompanied with changes in cell size and cell number. In shoots, contents of IAA, shikimates and proanthocyanidin were reduced, while contents of ABA, BL and leaf chlorophyll were increased. qRT-PCR revealed changes in multiple pathways of primary and secondary metabolisms. IAA treatment could recover many phenotypes and metabolisms of BnMYB69i plants. However, roots showed trends opposite to shoots in most cases, and BnMYB69i phenotypes were light-sensitive. Conclusively, BnMYB69s might be light-regulated positive regulators of shikimates-related metabolisms, and exert profound influences on various internal and external plant traits

    Optimized Dynamic Mode Decomposition via Non-Convex Regularization and Multiscale Permutation Entropy

    Dynamic mode decomposition (DMD) is essentially a hybrid algorithm based on mode decomposition and singular value decomposition, and it inevitably inherits the drawbacks of these two algorithms, including the selection strategy of truncated rank order and wanted mode components. A novel denoising and feature extraction algorithm for multi-component coupled noisy mechanical signals is proposed based on the standard DMD algorithm, which provides a new method solving the two intractable problems above. Firstly, a sparse optimization method of non-convex penalty function is adopted to determine the optimal dimensionality reduction space in the process of DMD, obtaining a series of optimal DMD modes. Then, multiscale permutation entropy calculation is performed to calculate the complexity of each DMD mode. Modes corresponding to the noise components are discarded by threshold technology, and we reconstruct the modes whose entropies are smaller than a threshold to recover the signal. By applying the algorithm to rolling bearing simulation signals and comparing with the result of wavelet transform, the effectiveness of the proposed method can be verified. Finally, the proposed method is applied to the experimental rolling bearing signals. Results demonstrated that the proposed approach has a good application prospect in noise reduction and fault feature extraction

    Wavelet analysis is a powerful tool for signal processing and mechanical equipment fault diagnosis due to the advantages of multiresolution analysis and excellent local characteristics in time-frequency domain. Wavelet total variation (WATV) was recently developed based on the traditional wavelet analysis method, which combines the advantages of wavelet-domain sparsity and total variation (TV) regularization. In order to guarantee the sparsity and the convexity of the total objective function, nonconvex penalty function is chosen as a new wavelet penalty function in WATV. The actual noise reduction effect of WATV method largely depends on the estimation of the noise signal variance. In this paper, an improved wavelet total variation (IWATV) denoising method was introduced. The local variance analysis on wavelet coefficients obtained from the wavelet decomposition of noisy signals is employed to estimate the noise variance so as to provide a scientific evaluation index. Through the analysis of the numerical simulation signal and real-word failure data, the results demonstrated that the IWATV method has obvious advantages over the traditional wavelet threshold denoising and total variation denoising method in the mechanical fault diagnose

    Improved Dynamic Mode Decomposition and Its Application to Fault Diagnosis of Rolling Bearing

    To solve the intractable problems of optimal rank truncation threshold and dominant modes selection strategy of the standard dynamic mode decomposition (DMD), an improved DMD algorithm is introduced in this paper. Distinct from the conventional methods, a convex optimization framework is introduced by applying a parameterized non-convex penalty function to obtain the optimal rank truncation number. This method is inspirited by the performance that it is more perfectible than other rank truncation methods in inhibiting noise disturbance. A hierarchical and multiresolution application similar to the process of wavelet packet decomposition in modes selection is presented so as to improve the algorithm’s performance. With the modes selection strategy, the frequency spectrum of the reconstruction signal is more readable and interference-free. The improved DMD algorithm successfully extracts the fault characteristics of rolling bearing fault signals when it is utilized for mechanical signal feature extraction. Results demonstrated that the proposed method has good application prospects in denoising and fault feature extraction for mechanical signals
